                实验原理

                ① 35 μL外泌体样品滴加在芯片上孵育;

                ② 预先包被的抗体特异结合外泌体Ψ表面蛋白以捕获外泌体;

                ③ 再使用荧光抗体特异性标记需要表征的标记物;

                ④ 用ExoView R100检测外泌体︽粒径、计数、蛋白表达(CD9,CD81,CD63等)及共定位。

                使用抗体Ψ捕获模式,防止样品中杂质影响结果,可直接检测血液、尿液和细胞培养液中的外泌体,未纯化样品的测量结果与纯△化后基本一致(如右图所示)。

                高精度SP-IRIS技术,可检测≥50 nm的外泌体,测量结果与电子显微←镜检测结果基本一致,并统计生成外泌体的粒径分¤布结果(如右图所示)。

                试剂盒配套◣相应的穿膜剂,可穿透外泌体并对外泌体内容物进行染色并检测,未穿膜时只能检测到跨膜蛋白CD9的荧光信号,穿膜后即可检测到外泌体内容物Syntenin的表达(如右图所示)。
